چکیده

'Wando ' pea is susceptible to Ditylenchus dipsaci f rom Raleigh, N. C. (RNC) but resistant to the same species from Waynesville, N. C. (WNC) . Homogenates of RNC and WNC were analyzed for pectolytic and cellulolytic enzyme activity; both had high Cx activity with W N C two to three times more active than RNC. Polymethylglacturonase activity was three to five times higher in RNC, but polygalacturonase was up to 100 times higher in WNC. Polygalacturonate-trans-eliminase was not detected although a Ca++-stimulated pectin methyl-trans-eliminase was present. Enzyme analyses of healthy and infected pea tissue showed only slight enzyme activity unrelated to that in nematode homogenates. No correlation between enzyme activity and the differing pathogenicities could be detected.

Role of nitrogen content of pea (Pisum sativum L.) on pea aphid (Acyrthosiphon pisum Harris) establishment

The leaf nitrogen content is generally accepted as an indicator of food quality and as a factor affecting host selection by phytophagous insects. The alate pea aphids (Acyrthosiphon pisum Harris, Aphididae) were given a choice among non-nodulated pea plants (Pisum sativum L.) supplied with one of four nitrate-N levels (0, 3, 15 and 30 mM). When whole plants were exposed to aphids for 7 days, th...

A Simplified Medium for Monoxenic Culture of Pratylenchus penetrans and Ditylenchus dipsaci.

P r e s e n t m e t h o d s o f establishing and m a i n t a i n i n g monoxenic cultures of plant parasitic nematodes are laborious. Use of complex, multicomponent nutrient media for production of callused host tissues is a factor c o n t r i b u t i n g to the inefficiency of these methods. For example, tile most commonly used of these media contains 19 constituents (1) . This n o t e r e p o...

Pectolytic enzymes in three populations of Ditylenchus dipsaci.

Extracts of nematodes of the Raleigh, North Carolina (RNC), Waynesville, N. C. (WNC), and onion populations of Ditylenchus dipsaci were examined for pectolytic activity. RNC nematodes contained a NaCl-stimulated endo-polymethylgalacturonase with optimal pH for activity of 6.0, whereas nematodes of the WNC and onion populations possessed a NaCl-stimulated endo-polygalacturonase with pH optimum o...
